For my personal favorite in a wheelgun: .41 Magnum
Why? Because it has plenty of power for almost anything I'm likely to shoot, can be handloaded from light to heavy loads, penetrates better than the .44 in many cases and has slightly less recoil.

What's that? Hard to conceal?
Not really.
At least this one isn't that difficult;
S&W Model 657, .41 Rem Magnum, 3-inch
For a semi-auto pistol, the .45 ACP still wins.
As much as the 10mm resembles the semi-auto version of the .41 Mag, most loadings - and most platforms - aren't using it to its full potential. They're slightly glorified .40 S&W loads.
The .45 ACP, however, still works well after almost 100 years of service.
