Stripped out the thread for a nipple in the back of my '51 Colt Navy. 
Any ideas how to fit it? Should I get a new cylinder?

If it stripped the cylinder instead of the nipple, I'd probably be considering Hawg's suggestion of drilling oversized and tapping for a bigger nipple.
At added expense, it may be possible to drill even farther oversize to permanently install a threaded plug. Then drill and tap the plug for the same size nipple.

personally I don't know of anybody that makes oversized nipples.
Although a sae thread nipple may be a bit bigger than the metric.
But you also need to consider the overall height of the nipple.
Best bet take it to machine shop, or gun smith have them install a helicoil restoring
it to original threads.
Or buy a new cylinder
This is another reason to remove the nipples once, coat with antisieze reinstall and then remove only once every few years.
You don't have to remove the nipples to make sure they are clean.
I haven'tremoved the nipples on my guns in 5 yr, but yet I know they will come out because of the coating, they always have in the past.

Just a guess, but
Are the threads for nipples exactly straight drilled and tapped relative to the cylinder body?
They might be at an angle.
A small one, but any angle will be a problem.
If so, it will take skill and a jig of sorts to do any replacement.
Might be worth a check before decided whether to just buy another cylinder.
Maybe 44 Dave has the best solution.
