Single action carry

Let me ask the firearms experts a question about my Colt Mustang Pocketlite 380.

Is it safe to carry with a round chambered, the hammer down, and safety on? The manual advises against this as pulling the trigger to lower the hammer disengages part of the internal safety mechanism, but...

I don't know if there's a real danger to this mode or if they're just being lawyer-shy.

Can you advise me?

No. Condition 2 carry, which you've described, bypasses every safety mechanism built into a Series 80 Colt and makes the weapons susceptible to firing if dropped. Don't do it.
