Can a S&W 625 handle the .45 Magnum?

Not advisable. The Smith .45 Colts have very thin chamber walls, especially at the cylinder stop notches.
I won't risk it in mine.

Probably not.

It may be able to handle the .460 Rowland though. The .460 Rowland is "juicer" than the .45 Win Mag.

You can call them @ 888 458 4126 if you're serious about wanting a more powerful round and don't mind all the extra cost/effort.
It is cheaper though than it would be to pick up a model 29 or 629 in .44 Rem Mag. - which the .460 Rowland pretty much equals.
 
I used to have a Hawes single action 45 Magnum. That was a monstrous revolver. Far larger and heavier than my Webley MKVI. I would not try 45 Magnum in a S&W 625 as it is a thinwalled cylinder. The 460 Rowland seems to be a way of shortening the lifespan of a 625. It is like juicing nitrous oxide in a Scion.
 
if you need a magnum buy a magnum. enjoy the 625 for what it is because it is a very fine shooting revolver
