Barrel length

Does the length of a 50 cal 1:28 twist 27 or 30 inch barrel really make a difference in accuracy. And if so how much?

If the longer barrel is more accurate, it's only because the increased sight radius makes it possible to aim more precisely.
With a scope, the short barrel may actually be more accurate.

Look up pictures of modern bench rest shooting, you don't see super long barrels on those rifles.
 
30"-32" for round ball have been found (per Wallace Gusler, late of Colonial Williamsburg) to be the most accurate. I wouldn't expect a 27" to be any significant difference.
