Anaconda/FA +P Durability

Looking for max penetration out of a .45 Colt. Looking at PMCs 300gr. +P FMJ, Grizzly 260gr +P JFN or 300gr +P FMJ. Suggestions?
Also, and most importantly, the hardware it'll be coming out of. Currently have the 6" .45 Anaconda. Will this safely handle such hot loads? I think it should, but want to draw on the more experienced for confirmation.
That said, Freedom Arms makes a .454Cassull/.45Colt with a 4.75" barrel. That would be much better for carry. I understand they also make one dedicated solely to .45Colt? What would be the advantage to this, weight? And how would it hold up to the aforementioned ammo?
I know Ruger is the established tough guy in this category, as apparently there is "Ruger Only" ammo. Just don't want one. And I want to make sure I'm not putting "RO" ammo thru the above.
Any feedback on the topic is very much appreciated

my Anaconda is a 6 inch barreled .44 magnum so based on that I'd look at comparable power loads in .45 Colt caliber for hunting and self defense, I'm not sure I'd plink with them though

The Anaconda is safe for published "Ruger-only" loads. The FA model 83 in .45Colt will handle loads that will fragment almost any other factory .45Colt. It is fully safe with custom five shot level loads in the 50-55,000psi range and beyond. Well beyond "Ruger only" territory. The factory .45 Redhawk is similar in strength. Some folks even believe that a five-shot .45Colt negates the need for the .454Casull. ;)

If what you want is penetration, skip the factory FMJ's and get a reloading setup. Cast bullets in the 335-360gr range will greatly outpenetrate any jacketed load.
